中文摘要:目的:探究人参花中内源激素分布规律,为后期研究内源性植物激素调控人参皂苷合成奠定基础。方法:采用超高效液相色谱-三重四级杆质谱(UPLC-MS/MS)对人参花中5种内源性植物激素(生长素,赤霉素,脱落酸,茉莉酸,水杨酸)含量进行测定分析。结果:人参花中5种内源性植物激素含量测定结果都具有专属性,与抗逆相关的激素脱落酸(ABA)和水杨酸(SA)含量较高,分别为1 126.17±232.52 ng·g-1和3 929.43±898.52 ng·g-1;与促生长相关的激素生长素(IBA)和赤霉素(GA3)含量较低,分别为516.70±138.55 ng·g-1和692.33±228.50 ng·g-1;茉莉酸(JA)合成途径中,茉莉酸前体12-氧植物-二烯酸(OPDA)含量极高,为47 158.84±12 910.55 ng·g-1;茉莉酸(JA)并未检出,茉莉酸活性形式异亮氨酸(JA-ile)为22.08±6.27 ng·g-1。结论:本研究首次检测了人参花中5种内源性植物激素的分布规律,并深入挖掘人参花中茉莉酸途径相关成分的合成,为后期深入研究内源性植物激素调控人参皂苷合成奠定基础。

Determination of Various Endogenous Hormones in Flower Buds of Panax ginseng by UPLC-MS/MS
Abstract:Objective:To explore the important regulation role of plant endogenous hormones on secondary metabolism of Chinese herbal medicine. Methods:Five kinds of endogenous phytohormones(auxin,gibberellin,abscisic acid,jasmonic acid,salicylic acid)in flowers of Panax ginseng were measured by UPLC-MS/MS.Results:The method for determination of 5 endogenous phytohormone content in P.ginseng all has high specificity.The higher content of ABA and SA(plant resistance related)was 1 126.17±232.52 ng·g-1 and 3 929.43±898.52 ng·g-1 respectively;The lower content of IBA and GA3(plant growth related)was 516.70±138.55 ng·g-1 and 692.33±228.50 ng·g-1 respectively;In jasmonic acid synthesis pathway,the highest content of jasmonate precursor,OPDA,was 47 158.84 + 12 910.55 ng·g-1.JA was not detected and JA-ile was 22.08+6.27 ng·g-1. Conclusion:The study explored the distribution of 5 endogenous phytohormones in ginseng flowers,which laid the foundation for further study on the regulation of endogenous phytohormones on ginsenoside biosynthesis.
