中文摘要:目的:构建四妙勇安汤活性部位配伍方薄层色谱鉴别方法和HPLC测定绿原酸和哈巴俄苷的含量测定方法。方法:采用薄层色谱法对配伍方中玄参、甘草进行鉴别,采用HPLC对配伍方中绿原酸和哈巴俄苷含量进行检测。结果:薄层色谱鉴别中配伍方中在与对照品哈巴俄苷、甘草酸铵相应位置上均显相同颜色的荧光斑点,斑点清晰,阴性对照无干扰。绿原酸和哈巴俄苷分别在0.195~1.56 0(r=0.999 9)、0.019 2~0.384 0 μg·μL-1(r=1.000 0)线性关系良好,平均加样回收率分别为96.5%、97.8%,RSD分别为0.9%、1.1%(n=9)。结论:薄层鉴别及绿原酸和哈巴俄苷的含量测定方法专属性强,准确稳定、重复性好,能够有效控制该配伍方的质量。

TLC Identification and Determination of Chlorogenic Acid and Habaroside in Active Part of Simiao Yongan Decoction
Abstract:Objective:To establish a TLC method for identification of active part and an HPLC method for determination of chlorogenic acid and habaroside in Si-Miao-Yong-An decoction by TLC.Methods:TLC was used to identify Radix Scrophulariae and Radix Glycyrrhizae,and HPLC was used to detect the content of chlorogenic acid and habaroside in decoction.Results:The fluorescence spots with decoction in the corresponding positions of Habaroside and ammonium glycyrrhizate were identified in TLC,and the spots were clear and the negative controls were not disturbed.Chlorogenic acid and habaroside had good linear relationship in the range of 0.195-1.560 μg·μL-1(r=0.999 9)and 0.019 2-0.384 0μg·μL-1(r=1.000 0)respectively,and the average recovery rate was 96.5%(n=9)and 97.8%(n=9),RSD was 0.9% and 1.1%,respectively.Conclusion:The TLC identification and determination of chlorogenic acid and habaroside are strong in specificity,accurate,stable and reproducible,and can effectively control the quality of the active part of Si Miao Yong An decoction.
