Identification of Flavonoids in Xiaoai Jiedu Decoction and Its Distribution in Rats
Abstract:Objective:Ultra-high performance liquid chromatography-tandem-quadrupole-time of fligh-mass spectrometry (UPLC-Q-TOF-MS) was used to identify the flavonoids in Xiaoai Jiedu Decoction (XJD) water extract and their distribution in rats after oral administration of XJD. Methods:The analytes were separated on an Agilent C18 column (100 mm×2.1 mm,1.8 μm),and eluted with a gradient of acetonitrile-water containing 0.1% formic acid. The mass spectrometer equipped with electron spray ionization (ESI) was used as detector and operated under the positive and negative ion mode. Results:23 flavonoids were isolated and identified. Among them,compounds 6-methoxynaringenin (13),naringenin (14),4′-hydroxwogonin (15),apigenin (16),wogonin (19) were detected in multiple biological samples. Among them,6,6,12,and 10 species flavonoids were detected in intestinal,brain,stomach,and stool samples,respectively. Conclusion:The liquid-mass method established in this experiment can quickly analyze and identify the flavonoids in XJD and the flavonoids in the effective parts of rats after oral administration of XJD.
