中文摘要:目的:采用高效液相色谱-四级杆/飞行时间串联质谱法(HPLC-Q/TOF-MS)建立苓桂术甘汤成分快速分析方法,实现对苓桂术甘汤物质基础的表征。方法:采用Welch Ultimate XB-C18色谱柱(250 mm×4.6 mm,5 μm),以流动相乙腈(B)-0.1%甲酸水(A)溶液进行梯度洗脱,流速为0.8 mL·min-1,柱温为25 ℃,进样量为10 μL。质谱采用电喷雾离子源(ESI)正、负离子模式,扫描范围mz 50~1000。结果:对正负模式下的质谱信息进行分析,并结合相关文献,共鉴定出苓桂术甘汤中60个化学成分,其中包括有机酸类成分5个、黄酮类成分17个、三萜类成分24个、内酯类成分5个,另有氨基酸类、核苷酸类、糖苷类等成分。结论:HPLC-Q/TOF-MS技术能实现对苓桂术甘汤的全成分快速分析、鉴定。

Rapid Analysis Components of Lingguizhugan Decoction with HPLC-Q/TOF-MS
Abstract:Objective:To establish a rapid analysis method by HPLC-Q/TOF-MS,and acquire the material basis characterization of Lingguizhugan Decoction. Methods:The analysis was performed on a Welch Ultimate XB-C18 (250 mm×4.6 mm,5 μm) HPLC column. The mobile phase consisted of acetonitrile (B) and 0.1% formic acid (A) was used as gradient elute. The flow rate was 0.8 mL· min-1,column temperature was 25 ℃,and injection volume was 10 μL. The ESI was operated in positive mode and negative mode,and scanning range was mz 50-1000. Results:Through the analysis of mass spectrometry data in positive and negative modes and with the help of relevant literature,a total of 60 chemical compounds in Lingguizhugan Decoction were identified,including 5 organic acid compounds,17 flavonoid compounds,24 triterpenoid compounds,5 lactone compounds,besides,amino acids,nucleotides,and glycosides were also identified. Conclusion:Via the HPLC-Q/TOF-MS technology,it was achieved to rapidly analyze and identify the whole compounds of Lingguizhugan Decoction.
