中文摘要:目的:建立忘忧安神方标准煎液(WAPSD)高效液相色谱法(HPLC)指纹图谱及4种指标性成分栀子苷、甘草苷、槲皮苷、甘草酸含量测定方法,为忘忧安神方(WAP)的质量控制提供参考。方法:制备10批WAPSD,采用高效液相色谱-二极管阵列检测器(HPLC-DAD)对WAPSD样品进行含量测定,建立 10 批WAPSD的HPLC指纹图谱,采用国家药典委员会“中药色谱指纹图谱相似度评价系统(2012版)”软件对其进行相似度评价,对共有峰进行归属及指认。结果:栀子苷质量分数为 36.84~55.86 mg·g-1,甘草苷质量分数为 3.96~6.57 mg·g-1,槲皮苷质量分数为4.59~13.76 mg·g-1,甘草酸质量分数为 16.48~21.40 mg·g-1,不同批次4种成分含量差异较大,说明不同批次的栀子、甘草、合欢花质量差异较大。建立了WAP的 HPLC 指纹图谱,10 批WAPSD指纹图谱与对照图谱相似度均大于0.94;标定了36个共有峰,多数峰来源于黄花菜、甘草、栀子;指认了4个共有峰,分别为17号峰栀子苷、24号峰苷草苷、26号峰槲皮苷、35号峰甘草酸。结论:所建立WAPSD多指标成分测定及指纹图谱方法灵敏度高、准确性好、有较高稳定性,能体现WAP组分的整体特征,可为WAP后续开发及其成品制剂的质量控制提供参考。

Establishment of HPLC Fingerprint and Determination Method for Four Index Components of Wangyou Anshen Prescription Standard Decoction
Abstract:Objective:To establish HPLC fingerprint of Wangyou Anshen Prescription standard decoction (WAPSD) and determine contents of four index components:geniposide,liquiritin,quercitrin and glycyrrhizic acid,so as to provide basis for quality control of Wangyou Anshen Prescription (WAP). Methods:10 Batches of WAPSD were prepared. The HPLC-DAD method was used for content determination and fingerprint of WAPSD,and the China Pharmacopoeia Committee ″2012 edition of Chinese Medicine Chromatographic Fingerprint Similarity Evaluation System″ software was adopted for similarity evaluation,and the common peak assignment and identification. Results:The contents of 10 batches of WAPSD were determined,among which the content of geniposide was 36.84-55.86 mg·g-1;the content of liquiritin was 3.96-6.57 mg·g-1;the content of quercitrin was 4.59-13.76 mg·g-1,and the content of glycyrrhizic acid was 16.48-21.40 mg·g-1,respectively. The contents of 4 components in different batches were significantly different,which indicated that the quality of the original medicinal materials in different batches was quite different. HPLC fingerprint of WAP was established,and the similarity between 10 batches of WAPSD fingerprint and common mode was greater than 0.94. Thirty-six common peaks were demarcated,most of which were derived from Hemerocallis Flava,Gardeniae Fructus and Glycyrrhizeae Radix,and four common peaks were identified as geniposide (peak 17),liquiritin(peak 24),quercitrin (peak 26) and glycyrrhizic acid (peak 35). Conclusion:The established WAPSD multi-index component determination method and fingerprint method have high sensitivity,good accuracy and high stability,which can reflect the overall characteristics of WAP components,and can provide the basis for the subsequent development of WAP and the quality control of its finished preparations.
