中文摘要:目的 建立高效液相色谱法(HPLC)同时测定杞菊地黄丸中莫诺苷、马钱苷、芍药苷、丹皮酚、绿原酸、木犀草苷、3,5-O-二咖啡酰基奎宁酸的含量。方法 采用Inertsil ODS-3 C18色谱柱(250 mm×4.6 mm,5 μm);以乙腈-甲醇-0.2%冰醋酸为流动相进行梯度洗脱;检测波长:240 nm(莫诺苷、马钱苷、芍药苷)、348 nm(绿原酸、木犀草苷、3,5-O-二咖啡酰基奎宁酸)、274 nm(丹皮酚);柱温为30 ℃;流速为0.9 mL·min–1;进样量为10 μL。结果 莫诺苷、马钱苷、芍药苷、丹皮酚、绿原酸、木犀草苷、3,5-O-二咖啡酰基奎宁酸分别在4.83~482.66(r=1.000 0)、5.02~501.95(r=1.000 0)、4.89~489.33(r=1.000 0)、10.38~207.63(r=1.000 0)、1.26~125.83(r=0.999 6)、0.63~62.98(r=0.999 7)、2.34~233.99 μg·mL–1r=0.999 8)线性关系良好,平均加样回收率分别为99.73%、101.00%、98.67%、99.19%、102.56%、101.66%、100.98%,RSD分别为1.29%、0.51%、1.21%、1.01%、0.97%、1.07%、0.47%。结论 该方法简便、快速、准确,可用于杞菊地黄丸的质量控制。

Simultaneous Determination of Seven Constituents in Qiju Dihaung Pills by HPLC
Abstract:Objective To establish a high performance liquid chromatography (HPLC) method for the simultaneous determination of morroniside, loganin, paeoniflorin, paeonol, chlorogenic acid, galuteolin, 3,5-O-dicaffeoylquinic acid in Qiju Dihuang Pills.Methods HPLC conditions are as below: Inertsil ODS-3 C18 column (250 mm×4.6 mm, 5 μm), gradient elution with acetonitrile-methanol-0.2% glacial acetic acid at the flow rate of 0.9 mL·min–1, column temperature of 30 ℃, injection volume of 10 μL, detection wavelengths of 240 nm (morroniside, loganin, paeoniflorin), 274 nm (paeonol), and 348 nm (chlorogenic acid, galuteolin, 3, 5-O-dicaffeoylquinic acid).Results Morroniside, loganin, paeoniflorin, paeonol, chlorogenic acid, galuteolin, and 3,5-O-dicaffeoylquinic acid showed good linear relationship within the concentration ranges of 4.83-482.66 (r=1.000 0), 5.02-501.95 (r=1.000 0), 4.89-489.33 (r=1.000 0), 10.38-207.63 (r=1.000 0), 1.26-125.83 (r=0.999 6), 0.63-62.98 (r=0.999 7), and 2.34-233.99 μg·mL-1 (r=0.999 8), respectively. The average recoveries were 99.73%, 101.00%, 98.67%, 99.19%, 102.56%, 101.66%, and 100.98%, with the RSDs of 1.29%, 0.51%, 1.21%, 1.01%, 0.97%, 1.07%, and 0.47%, respectively.Conclusion The method is simple, effective, and accurate, which can be used for quality control of Qiju Dihuang Pills.
