中文摘要:目的 优化补肾强筋丸的提取工艺。方法 采用高效液相色谱法,以地黄苷D、马钱苷、莫诺苷、阿魏酸含量和浸膏得率的总评归一值(OD值)为评价指标,以浸泡时间、提取时间、溶剂量为影响因素,应用星点设计-效应面法选取最佳提取工艺条件。结果 最佳提取工艺为12倍水提取2次,每次浸泡时间为60 min,提取时间为120 min,OD值为0.723 3。验证实验结果显示,实测值与预测值偏差为3.0%。结论 该方法稳定可行,可用于提取补肾强筋丸,为临床应用和新药的研发提供参考。 |

Extraction Process Optimization for Preparation of Bushen Qiangjin Pills by Overall Desirability and Response Surface Methodology |

Abstract:Objective To optimize the extraction process of Chinese medicinal materials for the preparation of Bushen Qiangjin pills.Methods High performance liquid chromatography was employed to determine the content of active components. The optimal extraction conditions were determined based on the overall desirability (OD) of rehmannioside D, loganin, morroniside, ferulic acid, and extract yield. The optimum soaking time, extraction time, and solvent volume were determined via central composite design-response surface methodology.Results The optimum extraction conditions: 12 volumes of water, extraction for twice, soaking time of 60 min, and extraction time of 120 min, under which the OD was 0.723 3. The results of validation test showed that the deviation between the predicted value and the measured value was 3.0%.Conclusion The optimized extraction conditions are stable and feasible and can be used for the preparation of Bushen Qiangjin pills, which provide reference for clinical application as well as the research and development of new drugs. |
