HPLC-DAD同时测定风热感冒颗粒中6种活性成分的含量

中文摘要:目的 建立HPLC-DAD同时测定风热感冒颗粒中苦杏仁苷、(R,S)-告依春、芦丁、连翘酯苷A、3,5-O-二咖啡酰基奎宁酸和牛蒡苷含量的方法。方法 采用液相色谱法,色谱柱为Waters SymmetryShieldTM RP18,流动相为乙腈(A)和0.1%的磷酸水溶液(B),梯度洗脱;流速:1.0mL.min-1;柱温:30℃;检测波长:苦杏仁苷和牛蒡苷为207nm,(R,S)-告依春245nm,芦丁、连翘酯苷A和3,5-O-二咖啡酰基奎宁酸为330nm。结果 苦杏仁苷、(R,S)-告依春、芦丁、连翘酯苷A、3,5-O-二咖啡酰基奎宁酸和牛蒡苷的线性范围分别为23~920μg.mL-1(r=0.9994), 0.4~16μg.mL-1(r=0.9995), 4.4~176μg.mL-1(r=0.9999), 2.8~110μg.mL-1(r=0.9998) ,5.4~216μg.mL-1(r=0.9996)和38~1520μg.mL-1 (r=0.9994),平均加样回收率(n=6)分别为98.8%、98.7%、98.4%、99.2%、99.8%和98.7%,RSD分别为1.0%、2.5%、1.8%、1.3%、1.3%和0.8%。结论 本法操作简便,结果准确,重现性好,可用于风热感冒颗粒的质量控制。

Simultaneous determination of six kinds of components in Fengreganmao Granules by HPLC-DADLiu Bin 1(1. Taian Testing Center for Food and Drug Control,Taian 271000)
Abstract:Objective To establish a HPLC -DADmethod for determination of amygdalin,(R,S)-epigoitrin, rutin , forsythiaside A, 3,5-O-dicaffeoyl quinic acid and arctiin in Fengreganmao granules. Methods A stable HPLC method was established and the chromatography was accomplished on a Waters SymmetryShieldTM RP18 column ,the mobile phase consisted of acetonitrile (A)-0.1% (w)phosphoric acid(B) as mobile phase in a gradient mode. The flow rate was 1.0 mL.min-1 and the column temperature was set at 30℃. The detection wavelength was set at 207nm for amygdalin and arctiin, 245 nm for (R,S)-epigoitrin,330nm for rutin , forsythiaside A and 3,5-O-dicaffeoyl quinic acid,respectively. Result The linear ranges of amygdalin,(R,S)-epigoitrin, rutin , forsythiaside A, 3,5-O-dicaffeoyl quinic acid and arctiin were 23~920μg.mL-1(r=0.9994), 0.4~16μg.mL-1(r=0.9995),4.4~176μg.mL-1(r=0.9999), 2.8~110μg.mL-1(r=0.9998),5.4~216μg.mL-1(r=0.9996) and 38~1520μg.mL-1 (r=0.9994),respectively.The average recovery were 98.8%,98.7%,98.4%,99.2%, 99.8% and 98.7%,RSD were 1.0%,2.5%,1.8%,1.3%,1.3% and 0.8%. Conclusion The method is simple to operate,accurate and reproducible, which can be used for the quality control of Fengreganmao granules.
