Abstract:[Abstract] Objective:To establish a research method of quality marker (Q-Marker) of goji berry based on UPLC fingerprint and antioxidant spectrum effect, and provide basis for its quality evaluation.Methods:UPLC was used to establish the fingerprints of 31batches of goji berry.Based on the results of the common peak areas of the fingerprint, the quality evaluation and screening the main quality markers of goji berry was using clustering analysis, principal component analysis, orthogonal partial least squares discriminant analysis.The antioxidant capacity of goji berry was evaluated by the determination of MFO, DPPH and T-AOC. Bivariate correlation analysis and partial least squares regression were used to analyze the spectral - effect relationship between common peaks and antioxidant activity indexes.Comprehensive analysis and screening of quality markers.Results:The fingerprints identified 34 common peaks. 31 batches of goji berry were divided into 3 categories, which could distinguish different origin of goji berry.Principal component analysis showed four main factors affecting the classification of goji berry. Orthogonal partial least squares discriminant analysis showed that the compound with common peaks 2, 7, 17, 19 and 33 could be used as the main quality markers for the identification and differentiation of goji berry.Bivariate correlation analysis showed that compounds with common peaks 8, 11, 15, 20, 23 and 34 were significantly correlated with the values of MFO, DPPH and T-AOC. Partial least squares regression analysis showed that compounds with common peaks 11, 15, 23, 25, 31, 33 and 34 were the main active ingredient groups,these compounds can be used as quality markers of antioxidant activity of goji berry.Conclusion:This study can clarify the material basis of antioxidant effect of goji berry ,the quality markers could provide a theoretical basis for quality control and evaluation of goji berry. |
