中文摘要:目的:将发酵虫草菌粉(Cs-4)粗多糖进行分离纯化,并观察纯化产物对ConA抑制人正常肝细胞(L-O2细胞株)增殖活性的拮抗作用。方法:发酵虫草菌粉(Cs-4)通过水提醇沉、复溶、透析、浓缩、冷冻干燥得粗多糖,采用DEAE-纤维素离子交换、sephadex-G-100柱色谱分离纯化粗多糖,MTT法检测细胞增殖活性。结果:发酵虫草菌粉(Cs-4)粗多糖经分离纯化得到均一成分,命名为CSMP-60-B-I。高效凝胶渗透色谱法测得其相对峰高分子量为32 336 Da。毛细管电泳测得单糖组成为木糖-阿拉伯糖-葡萄糖-半乳糖-甘露糖(0.09∶0.20∶1.00∶3.70∶3.58)。高浓度ConA能抑制L-O2细胞增殖,而加入CSMP-60-B-I后,这种抑制作用大大减弱。结论:发酵虫草菌粉(Cs-4)粗多糖的分离纯化产物CSMP-60-B-I对ConA抑制L-O2细胞增殖活性有拮抗作用,提示其可能有保肝作用。

Physicochemical Properties of Homogeneous Polysaccharide Extracted from Fermented Cordyceps Powder and It’s Antagonism of Inhibition Effect on Cell Proliferation Activity of L-O2
Abstract:Objective:To isolate and purify the total polysaccharides extracted from fermented Cordyceps powder(Cs-4)and observe the antagonism of inhibition effect on cell proliferation activity of L-O2.Methods:The fermented Cordyceps powder(Cs-4)was extracted with water at 60 ℃ and precipitated with ethanol,then redissolved,dialyzed,concentrated and lyophilized to obtain the total polysaccharides.The total polysaccharides were fractionated by a DEAE-cellulose column.The cell proliferation activity of L-O2 was detected by MTT.Results:The total polysaccharides extracted from fermented Cordyceps powder(Cs-4)were fractionated to obtain homogeneous polysaccharide named CSMP-60-B-I.The relative peak molecular weight of CSMP-60-B-I was 32 336 Da,determined by high performance gel permeation chromatography.The composition of monosaccharides and molar ratios were xylose-arabinose-glucose-galactose-mannose(0.09:0.20:1.00:3.70:3.58),determined by capillary electrophoresis.The results showed that ConA in high concentration could inhibit the cell proliferation activity of L-O2 in vitro.After the addition of CSMP-60-B-I,the inhibiting effect was greatly reduced.Conclusion:CSMP-60-B-I may have the effect of reducing inhibition of cell proliferation activity of L-O2by ConA in high concentration,implicating that CSMP-60-B-I might have hepatoprotective effect.
