中文摘要:目的:优化裸花紫珠茎叶水提取的工艺及初步考察水提取物的外用止血作用。方法:以浸膏得率和毛蕊花糖苷得率为评价指标,考察浸泡时间、加水倍量、提取时间和提取次数等因素,采用正交试验优选最佳水提取工艺。采用家兔耳缘静脉出血模型,外用0.5、1.0、2.0 g·mL-1(按生药量计)三种浓度的水提取物,以出血量和出血时间为指标,评价裸花紫珠的外用止血效果。结果:裸花紫珠最佳水提取工艺为浸泡时间2 h,提取次数3次,加水倍量为12、10、10,每次提取时间为1 h;与空白组相比,裸花紫珠低、中、高剂量组出血量均显著减少(P<0.01),并且高剂量组相较空白组出血时间显著缩短(P<0.01)。结论:优选出的水提取工艺简单可行,且裸花紫珠水提取物有一定的外用止血作用,有进一步开发为外用止血贴的潜力。

Optimization of Water Extraction Process of Callicarpa nudiflora by Orthogonal Design and Evaluation of Hemostasis Effects in External Use
Abstract:Objective:To optimize the water extraction process of stem and leaves of Callicarpa nudiflora and preliminarily evaluate hemostasis effects of water extract in external use.Methods:The optimum extraction process was optimized by orthogonal experiment with the yield of extraction and the extraction rate of acteoside as evaluation indicator.The influence of soak time,water addition,extraction time,and extraction time were investigated.The bleeding wound model of rabbit ear vessels was established and three concentrations of water extract (0.5,1.0,2.0 g·mL-1,in terms of crude drug) were external used respectively.The hemostasis effects were evaluated with the bleeding volume and the bleeding time as evaluation indexes.Results:The optimized water extract craft of C.nudiflora was:the soak in water for 2 h,the extraction time was 3,with 12,10,10 times water in sequence,and each extraction time was 1h.Compared with control group,the amount of bleeding decreased significantly in each dose group (P<0.01),and bleeding time also decreased significantly in high dose group (P<0.01).Conclusion:The optimized extraction process is simple and practicable,the water extract of C.nudiflora has certain hemostasis activity in external use and there is potential for further development of external hemostatic patches.
