中文摘要:目的:优化夏至草总黄酮的超声提取工艺,建立夏至草总黄酮的含量测定方法。方法:在单因素试验的基础上,采用Box-Behnken设计结合响应面分析法,以夏至草总黄酮的提取率为评价指标,采用分光光度法,测定夏至草中总黄酮的含量,采用Design-Expert v8.0.6软件对试验结果进行拟合和建模,确定夏至草总黄酮超声提取的最佳工艺。结果:超声法提取夏至草总黄酮的最佳工艺为乙醇浓度60%,液料比40∶1,温度60 ℃,在此条件下,夏至草总黄酮提取率为1.592 4%(RSD=1.007 7%,n=6)。结论:该方法操作简便、准确、灵敏、重现性好,可作为夏至草总黄酮的提取及含量测定方法。 |

Optimization for Extraction Process of Total flavonoids from Lagopsis supina by Response Surface Methodology |

Abstract:Objective:To optimize the extraction of total flavonoids of Lagopsis supina and establish a method for determination of the total flavonoids content.Methods:Using the extraction yield of total saponins as index,spectrophotometric method was used to determine the content of total flavonoids in L.supina.On the basis of single factor experiments,Box-Behnken design and response surface were used to optimize the extraction of total flavonoids.And the extraction yield of the total flavonoids was analyzed using quadratic regression model by the software of Design-Expert v8.0.6.Results:The optimum extraction conditions were ethanol concentration 60 %,liquid to material ratio 40∶1,temperature 65 ℃.Under these conditions,the yield of extracted total flavonoids could reached to 1.592 4% (RSD=1.007 7%,n=6).Conclusion:The method is simple,accurate,sensitive and reproducible,and can be used for content determination of total flavonoids from L.supina. |
