中文摘要:目的:测定虎杖不同根部位(根木质部、根茎木质部、根表皮、根茎表皮、根、根茎)中的5种活性成分含量,为虎杖的综合利用和质量控制提供科学依据。方法:色谱柱为Agilent Extend C18柱(250 mm×4.6 mm,5 μm),流动相为乙腈和水,柱温为40 ℃,检测波长为290 nm,流速为1.0 mL·min-1。结果:5种成分线性良好(r>0.999),平均加样回收率为98.4%~98.8%。虎杖苷在根茎表皮中含量最高,为72.48 mg·g-1;白藜芦醇和大黄素在根表皮中含量最高,分别为8.83 mg·g-1和23.96 mg·g-1;大黄素-8-O-β-D-葡萄糖苷在根茎木质部中含量最高,为40.57 mg·g-1;大黄素甲醚在根茎木质部中含量最高,为17.24 mg·g-1。结论:该方法快速、准确,操作简单,专属性强,可为虎杖的质量评价提供参考依据。

Simultaneous Determination of Five Active Components in Different Parts of Polygonum Cuspidatum Root by HPLC
Abstract:Objective:The contents of five active ingredients in different parts of Polygonum cuspidatum (root xylem,rhizome xylem,root epidermis,rhizome epidermis,root,rhizome) were determined,providing scientific basis for the comprehensive utilization and quality control of Polygonum cuspidatum.Methods:The chromatographic column was Agilent Extend C18 column (250 mm×4.6 mm,5 μm),the mobile phase was acetonitrile and water,the column temperature was 40 ℃,the detection wavelength was 290 nm,and the flow rate was 1.0 mL·min-1.Results:The five components had a good linearity (r>0.99),with an average recovery rate of 98.4%-98.8%.The highest content of polydatin in rhizome epidermis was 72.48 mg·g-1,the highest content of resveratrol and emodin was 8.83 mg·g-1 and 23.96 mg·g-1 respectively,the highest content of emodin-8-O-β-D-glucoside in rhizome xylem was 40.57 mg·g-1,and the highest content of physcion in rhizome xylem was 17.24 mg·g-1.Conclusion:The established method is rapid,accurate,simple and specific,and can provide reference for the quality evaluation of Polygoni Cuspidati Rhizoma et Radix decoction pieces.
