天麻萌发菌胞外酶活性变化规律研究

中文摘要:目的:研究天麻萌发菌03、06菌株不同时期的胞外酶活性变化规律及菌株生长速度,为天麻有性繁殖优良萌发菌菌株筛选提供理论依据。方法:利用木屑-麦麸培养基培养菌株03和06,测量生长量,采用2,2-联氮-二(3-乙基-苯并噻唑-6-磺酸)二铵盐(ABTS)法和3,5-二硝基水杨酸(DNS)法测定不同生长时期2种菌株胞外酶活性。结果:培养前期2种菌株生长速度逐渐增加,03菌株生长速度大于06菌株,后期2种菌株生长速度有所减缓。漆酶和木聚糖酶是天麻萌发菌生长过程中活性较强的胞外酶,06菌株木聚糖酶活性和淀粉酶活性均高于03菌株。结论:通过对2种菌株生长速度、胞外酶活性的比较和分析,初步认为03为优势菌株。

Study on Activity Changes of Extracellular Enzyme of Gastrodia elata Germinating Fungi
Abstract:Objective:To provide theoretical reference for selecting fine strains in the sexual reproduction of Gastrodia elata by studying the activity changes of extracellular enzymes during the growth and the rate of two G. elata germinating fungus strains. Methods:The two fungus strains 03 and 06 were cultivated with wood chip-wheat bran medium,the increment and the activities of extracellular enzymes were measured at different stages by adopting ABTS and DNS. Results:The growth rate of the two strains increased gradually in the early stage of cultivation,and the growth rate of the 03 strain was faster than that of the 06 strain. The growth rate of the two strains slowed down in the later stage. Laccase and xylanase were the active extracellular enzymes during the growth of strains. The activities of xylanase and amylase of the 06 strain were higher than those of the 03 strain. Conclusion:By comparing and analyzing the growth rate and extracellular enzymes activities of the two strains,it can be considered that the 03 strain is the dominant strain for Gastrodia elata germinating.
